Duke has an older sister named Cindy and does not have a relationship with his father. At the age of nine, his mother moved the family to Brooklyn, New York City, where they settled in a studio apartment. This move was motivated by the desire for better educational opportunities for his sister, who was pursuing medical school. Duke attended a public primary school in Crown Heights and often frequented the local library and comic book store after school. He graduated from Brighton High School in Rochester, New York, in 2004. He then attended the University at Buffalo, where he earned a Bachelor of Arts in theater, followed by a Master of Fine Arts in acting from Yale University, graduating in 2013.
